ChristianaCare is searching for a Research Scientist II to focus on Oncology Gene Therapy.

Non-small cell lung cancer (NSCLC) is the leading cause of cancer-related death worldwide and in the United States. There is a high unmet need for new treatment options to treat NSCLC due to the high rate of chemoresistance in NSCLC. Gene Editing Institute at Christiana Care Health System has identified a specific target to reduce chemoresistance and is currently developing a CRISPR gene therapy strategy to treat NSCLC. This platform protocol is also applicable to other cancers.  

The Research Scientist will participate in all aspects of preclinical development work in a new, targeted delivery system utilizing CRISPR gene therapy to treat NSCLC. The successful candidate will support the clinical development of drug pipelines by designing and executing in vitro and in vivo experiments, developing protocols and standards, and mentoring junior staff scientists. The Research Scientist is also involved in the development of discovery, proof-of-concept, and preclinical studies for other solid cancers. The ideal candidate will be part of a team that leads a transformative advance in patient care.

PRINCIPAL DUTIES AND RESPONSIBILITIES:

  • Supervise and perform hands-on in vivo animal experiments including designing the studies, developing and maintaining subcutaneous and orthotopic xenografts in mice, and carrying out various delivery modes of vectors and drugs.

  • Establish and perform quantitative assessment workflows by standardizing protocols for ddPCR, qPCR, qRT-PCR, plate-based assays, and quantitative western.

  • Engineer gene therapy products using site-specific genome editing in lung cancer models (designing & using appropriate genome editing techniques including viral vector transduction).

  • Coordinate and participate in viral vector design and construction

  • Explore new drug targets and lead and develop preclinical studies.

  • Work with Contract Research Organizations to conduct tropism, efficiency analysis, biodistribution and toxicology studies for timely execution of projects.

  • Present research findings, including preparation of research reports and abstracts, dissemination at national and international scientific conferences and publication of manuscripts by peer-reviewed scientific journals. 

  • Keep up with current scientific literature and techniques in the field of gene therapy.

EDUCATION AND EXPERIENCE REQUIREMENTS:

  • PhD in oncology, molecular biology, cell biology, biological engineering, biomedical engineering, or a closely related field.

  • Minimum of 4 years of industry experience or 5 years of postdoctoral experience in biopharmaceutical, biotechnology industry, or translational academic environment focusing on developing gene therapy products.

  • Candidates should also have authored at least two first author publications in reputable journals; cancer research experience preferred.

KNOWLEDGE, SKILL, AND ABILITY REQUIREMENTS:

  • Proficiency and extensive experience in xenograft animal models for gene therapy.

  • Proven ability to develop protocols and streamline quantitative analysis including ddPCR, qPCR, qRTPCR, and western.

  • Deep understanding of site-specific genome engineering technologies with extensive experience in the development of gene therapy products.

  • Experience in delivery systems including AAV, Ad5, and LNP, and viral vector designs.

  • Ability to work on mammalian cell cultures, patient-derived cells, flow cytometry, IHC, IF

  • Experience in preclinical research in biotech setting is preferred.

  • Strong leadership and communication skills

  • Evidence of scientific accomplishments in the form of publications and presentations as first author

  • Ability to work independently to develop model concepts and approaches for translational cancer research.

  • Extensive experience in research methodology, principles, and procedures

  • Ability to gather and analyze statistical data and generate reports.

Supervise junior staff and oversee daily laboratory operations.
